Application of the nourseothricin acetyltransferase gene (nat1) as dominant marker for the transformation of filamentous fungi

چکیده
Here, we report the construction of two transformation vectors, pD-NAT1 and pG-NAT1, carrying the nat1 gene encoding the nourseothricin acetyltransferase. The nat1 gene is expressed under the control of the Aspergillus nidulans trpC promoter and thus can be used as a dominant drug-resistance marker for the DNAmediated transformation of filamentous fungi. The successful application of both vectors was demonstrated by transforming the homothallic ascomycete Sordaria macrospora as well as the β-lactam producerAcremonium chrysogenum. For both fungi and for both vectors, transformation frequencies were between 10 and 40 transformants per 10 μg of plasmid DNA. Nourseothricin acetyltransferease: a positive selectable marker for Toxoplasma gondii.
Molecular analysis of parasite genomes will require new molecular genetic tools. The nat1 gene of Streptomyces noursei encodes nourseothricin acetyltransferase, conferring resistance to the aminoglycoside antibiotic nourseothricin. Electroporation of nat1 cassettes into RH or Prugniaud strains of Toxoplasma gondii allows for selection of stable nourseothricin-resistant clones.
متن کاملBiotransformation of Progesterone by Whole Cells of Filamentous Fungi Aspergillus brasiliensis
Microbial steroid biotransformation have found wide-reaching application for the production of more precious and functionalized compounds due to their high regio-and stereoselectivity. In this study, the possibility of using filamentous fungi Aspergillus brasiliensis cells in the biotransformation of progesterone (I), a C-21 steroid hormone was studied for the first time. The fungal strain was i...

متن کاملTransformation of Aspergillus sp. and Trichoderma reesei using the pyrithiamine resistance gene (ptrA) of Aspergillus oryzae.
A pyrithiamine (PT) resistance gene (ptrA) was cloned from a PT resistant mutant of Aspergillus oryzae and was useful as a dominant selectable marker for transformation of all A. oryzae wild type strain as well as A. nidulans. For further study, we examined whether or not ptrA could be used as the transformation marker in other species of filamentous fungi. Two types of plasmid, which contain p...
متن کاملDeprenyl increases synaptophysin and choline acetyltransferase in rat after sciatic nerve axotomy
Neuroprotective effect of deprenyl on motoneurons of spinal cord after axotomy of peripheral nerves such as sciatic has been well established. Deprenyl is an inhibitor of monoamine oxidase type-B (MAO-B). The main function of this agent is the release of neurotransmitters from pre-synaptic terminals. Acetylcholine is a neurotransmitter that is synthesized by choline acetyltransferase (ChAT) and...
